The general goal was to provide designs which eliminated bias and reduced random error. There are extensive sections on complex combinatorial designs such as Latin squares and partially replicated factorial designs, but there are also crucial basic sections on the importance of randomization and a discussion of the ability of replication to minimize random errors. Sections 9–11 on randomization and replication are fundamental and chapter IV (sections 22–30) which describes the randomized block design are particularly relevant.

The need for large numbers of patients to detect moderate treatment differences led to the development of the overview of all trials addressing the same question. The importance of collecting and updating follow-up on an individual patient basis was first demonstrated by Cuzick et al. (Paper 9) and led to important changes in the use of radiotherapy. Larger efforts, looking at all aspects of breast cancer treatment, were subsequently undertaken by the Early Breast Cancer Trialists Collaborative Group and an important exposition of overview methodology was published in one of their early reports (Paper 12).
